Serial sections stained for ATPase activity at pH 4.2 and pH 10.3. To distinguish type I and type II muscle fibres. Type I skeletal myofibres are also called “slow twitch,” they are responsible for long-duration, low-intensity activity. Type II fibres are also called “fast twitch,” have less myoglobin and are responsible for short-duration, high-intensity activity and quick bursts of energy. They can be discerned by ATPase activity.
Consecutive sections:
(A): pH 4.2, (1) is type I positive. (2) type II is negative.
(B): pH 10.3, (2) type II is positive. (1) type I is negative
