Title: Vagina (human, adult)
Description: Stain: (A) Azan; (B) Periodic acid Schiff (PAS).
(A): Non-keratinized stratified squamous epithelium (NKSSE) sharply demarcated by a blue-stained basement membrane (→) from the proper lamina (2). The NKSSE is composed of basal cells (3), parabasal cells (4) and transparent swollen intermediate and superficial cells (1) (due to a high glycogen content).
(B): Another similar specimen demonstrates reddish-stained glycogen in the upper cell layers (5) of the NKSSE (due to methanol-containing fixatives part of the glycogen is dissolved and the remnants are pushed basally). The parabasal cells (4) contain faintly dots of glycogen, (3) basal cells on a PAS-positive basement membrane, (2) proper lamina. Note intraepithelial lymphocytes (→).
