Title: Papillary serous cystadenocarcinoma of ovary (human, adult)
Description: Case nr. p577. Stain: (A) Hematoxylin-eosin (Weigert); (B) Immunoperoxidase staining with diaminobenzidin reaction (DAB) and hematoxylin counterstaining on frozen sections with OC125 antibody; (C) and OVTL3 antibody.
(A): Invasion of papillary adenocarcinoma. Solid sheets of small tumor cells (1) accompanied by acinar formation of larger neoplastic cells (2).The latter show distinct nuclear atypia, multinucleation, presence of scattered nuclear hyperchromatism and mitoses.
(B): Most tumor cells (3) express brown-stained membrane-bound OC125 reactivity. Stroma is negative (4).
(C): All tumor cells (3) including mitotic ones stain yellow-brown positive with OVTL3 antibody that also appears membrane-bound. Stroma is negative (4).
Background: See also Background POJA-L1476+L1760+L1770+L1750. Solid areas of the tumor are composed of closely packed papillae. Destructive infiltrative stromal invasion is evident as the tumor ingrowth is diffuse or as solid cell sheets. Extreme degrees of nuclear atypia indicate progress toward undifferentiated tumor cells. The OC125 (MUC16) antibody recognizes an epitope on a molecule called Cancer Antigen 125 (CA125) a.o. in tumor cell membranes. It is directed against a glycoprotein produced by human epithelial ovarian cancer cells. The antigenic determinant CA125 is expressed on more than 80% of all non-mucinous epithelial ovarian tumors. It does not react with mucinous ovarian tumors. It is not a tumor-specific antigen, its presence was also demonstrated in fetal as well as in adult normal tissues derived from coelomic epithelium. The antibody OVTL3 was generated against ovarian carcinoma cells and detect for the greater part cell membrane-bound epitopes of the carcinoma cells in the primary tumor as well as in distant metastases. The antibody was also applied for immunoscintigraphy.
