Title: Follicles and antrum formation in ovary (rabbit, human)
Description: (A) Scheme (human). Stain: (B,C) Azan (rabbit); (D) Hematoxylin-eosin (human); (E) Keratin 5,7,14,19 antikeratin antibody (OVTL12/5) immunoperoxidase staining with diaminobenzidin reaction (DAB) and hematoxylin counterstaining (human). Note: the individual magnifications in this photomontage are irrelevant.
(A): Scheme (human): antral follicle (tertiary follicle early type 5) with more than five layers of granulosa cells and a cavity. Cell-rich ovarian stroma of cortex (1); external theca with more fibrous components (collagen) (2); internal theca with differentiation of stromal cells into spindle-shaped cells (3); mural granulosa cells (4); follicular theca (i.e. theca folliculi) as a bilayered stromal sheath (5); antrum folliculi with liquor folliculi (6); corona radiata (ring of granulosa cells surrounding oocyte within the cumulus) (7).
(B): Rabbit: transition to antral follicle (secondary follicle late type 4) (a4), multilaminar secondary follicles (type 3 and type 4) (s3, s4) and proliferating primary follicle (type 2) (p2).
(C): Rabbit: secondary follicle late type 4 (transition to antral stage) more than five layers of granulosa cells and a distinct formation of a cavity (transversal section) (6). Oocyte with vesicular nucleus and nucleolus; a bluish-stained zona pellucida surrounded with a corona radiata (7, ring of granulosa on top of the cumulus). Mural granulosa cells (4), internal theca cells and blood vessels (3), external theca with blue-stained collagen fibers (2).
(D): Human: tertiary follicle early type 5 (antral stage) undergoing early atresia. Signs of degenerating oocyte (7) and start of collapsing zona pellucida. Cumulus and mural granulosa cells are less compact with interspersed few Call-Exner bodies (→). Internal theca, blood vessels, fibrous external theca, cell-rich stroma.
(E): Human: secondary follicle late type 4 (transition to antral stage). Broad spectrum cytokeratin (OVTL12/5) shows positive staining for granulosa cells of the large as well as in tangentially cut smaller follicles.
Background: Unilaminar primary follicle type 2 (preantral stage) shows a single layer of cuboidal granulosa cells. Proliferating primary follicle shows partially a second layer. Secondary follicle type 3 (preantral stage) has two-four layers of granulosa cells. Secondary follicle type 4 (preantral stage) is multilaminar and has four-six layers of granulosa cells and starts forming an intercellular cavity as well as a theca layer. Late type 4 is the transition stage to early antral stage with increased amounts of granulosa cells and formation of an antrum folliculi. Tertiary follicle type 5 (antral stage) with more than five layers of granulosa cells and an antrum folliculi.
