Description: Stain: (A) Hematoxylin-eosin.
(A): The colon (A) is composed of the following layers:
(1): Crypts lined with simple columnar resorptive epithelium and goblet cells.
(2): A loose connective tissue, the so called lamina propria infiltrated with lymphoid cells.
(3): The lamina muscularis mucosae.
(4): Tela submucosa.
(5a, b, c): The tunica muscularis with three bands (taeniae) of smooth muscles.
(B): SDH positive in the colon crypts, being related to the water resorption activity of the epithelial cells.
(C): ZFFA-positive in the surface epithelial cells and the upper crypts.
(D): Gpox with a similar pattern as the SDH.
(E): AFFA with positive reaction in the lamina muscularis mucosae and just one layer of the tunica muscularis mucosae.
(F): Note that all crypts are stained strongly positive in the PAS-reaction predominantly because of the goblet cells producing mucus. The matrix around the muscle fibers is also PAS.
